CLONING A FOREIGN GENE IN THE YEAST PICHIA PASTORIS

Abstract. The S gene encoding the hepatitis B surface antigen was cloned into the yeast Pichia pastoris using the pPIC9 transfer vector. The recombinant plasmid pPIC9-S was treated with the restriction enzyme SacI and inserted into the yeast Pichia pastoris via electroporation. Yeast clones were selected by growing in a medium without histidine. As a result, Mut+ phenotypes of Pichia pastoris containing the S gene were obtained.
